浙江省农业科学院林福呈团队综述线粒体自噬在植物-病原真菌生物互作中的作用

近日,林福呈团队以浙江省农科院(国重室)为第一通讯单位在国际自噬研究领域顶级期刊AutophagyIF5y=16.142)上发表题为“Current opinions on mitophagy in fungi”的综述论文。该文章第一作者为沈自芳博士,通讯作者为林福呈教授,浙江省农科院李琳博士、朱学明博士,浙江大学刘小红教授和美国密西根大学Klionsky教授参与了文章的撰写与修改。该论文得到了国家自然科学基金和浙江省自然科学基金,农产品质量安全危害因子与风险防控国家重点实验室等经费的支持。

线粒体自噬是确保线粒体质量控制的最重要的细胞过程之一,其目的是将受损、老化、功能失调或过剩的线粒体运输到液泡(植物和真菌)或溶酶体(哺乳动物)中进行降解和再利用。近年来的研究表明,线粒体自噬在病原真菌的产孢、营养生长和毒力中起着至关重要的作用,这与哺乳动物和酵母中的作用有很大的不同。文章总结展望了线粒体自噬在病原真菌中的研究现状,这将有助于研究者和读者更好地了解线粒体自噬的多重功能,并为研究线粒体自噬在真菌发病机制中的作用提供新的视角。

线粒体自噬发生的前提是待降解线粒体的准确识别。如图1所示,生物体中线粒体自噬的发生可以分别五个步骤,分别为(1)线粒体自噬受体的激活;(2)受体通过其AIM基序与核心自噬蛋白Atg8互作,招募自噬膜;(3)待降解的线粒体被自噬小体精确包裹;(4)自噬小体与液泡或溶酶体融合;(5)过量和受损的线粒体被精准降解,分解产物被释放回细胞质中再利用。

目前的研究表明,受体介导的线粒体自噬在真核生物中普遍保守,所有已发现的选择性自噬受体均存在能够与Atg8相互作用的AIM基序,该的特征是含有W/F/Y-X-X-L/I/V序列,其中X可以是任何酸性氨基酸(图2)。此外,线粒体自噬还与翻译后修饰过程磷酸化、线粒体分裂和融合以及线粒体呼吸链复合体III密切相关。多项证据表明磷酸化是受体介导的线粒体自噬的一个关键事件,病原真菌侵染菌丝在植物细胞中的扩展依赖于线粒体点状和丝状的形态变化。

稻瘟病严重威胁全球水稻产量和粮食安全,自噬调控稻瘟病菌致病性的机理研究是国际研究热点。30年来,林福呈教授及其团队一直致力于稻瘟病菌致病机理和绿色防控研究,取得了国内外公认的研究成果,在Molecular Plant AutophagyNew Phytologist mBio等高水平期刊上发表了158篇有影响力的SCI论文,他引10000余次。近年来团队积极参与湘湖实验室的谋划和启动建设,在农业生物互作领域的基础理论和关键技术研究方面双向发力。20203月份至今,团队发表5分以上高水平文章37篇,10分以上高水平文章6篇;授权国家发明专利5项,鉴定成果1项,申请国际专利5项,为浙江坤伦有机肥料有限公司带来近1.9亿的利润。
